Q: Does System Center Virtual Machine Manager 2012 need a VMware vCenter instance deployed to manage VMware ESX?

SCVMM needs vCenter. Desperately.

A: VMware doesn't expose any APIs for direct communication to ESX, so all management has to go through vCenter, which has public APIs. Therefore, System Center Virtual Machine Manager still requires vCenter to be deployed to manage ESX hosts.
